The overall aim of this project is to identify the regulatory network which controls constitutive and regenerative neurogenesis. This network involves transcription factors, enhancers and short non coding RNAs like enhancer RNA (eRNA) and micro RNA (miRNA). ERNAs are short RNA molecules that indicate the activity of an enhancer. MiRNAs are RNA molecules which regulate transcription of their target mRNA by sterically blocking or degrading the mRNA. After identification of different miRNA and eRNAs via RNA sequencing we want to validate the candidates and corresponding targets by different techniques like in situ hybridization or gene knockout. However, both miRNAs and eRNAs are difficult to detect because of low abundance and short nucleotide sequences. Therefore, for better visualization we want to develop synthetic fluorescent probes with high sensitivity and specificity. Zebrafish is used as a model organism in this project, since it has the remarkable ability to regenerate neurons even in adult stages and is therefore suitable to study neurogenesis.
